The basic aim of prodrug design is to mask undesirable drug properties, such as low solubility in water or lipid membranes, low target selectivity, chemical instability, undesirable taste, irritation or pain after local administration, presystemic metabolism, and toxicity.
The pharmacokinetic studies provide valuable information regarding the in vivo properties of a drug's limitations, such as poor absorp- tion, too rapid elimination, and presystemic metabolism.
It facilitates in several ways: improvement in absorption (by oral and nonoral routes), decreasing presystemic metabolism to improve time profile, improvements due to change in formulation, and increase in organ/tissue-selective delivery of the active agent.
